Abstract: This dissertation contains results of the studies relating to fabrication of the label free and, novel nanostructured immunosensor platform (BSA/anti-CEA/PEDOT/RGO/ITO) for efficient detection of a cancer biomarker, carcinoembryonic antigen (CEA). Nanostructured PEDOT/RGO has been synthesized via one step low temperature hydrothermal process and surface modified by covalent immobilization of monoclonal anti-CEA. Structural and morphological characterization of the fabricated electrodes has been carried out using X-ray diffraction (XRD), transmission electron microscopy (TEM) and scanning electron microscopy. The results of the response studies reveal that the BSA/anti-CEA/PEDOT/RGO/ITO immunoelectrode can be used to estimate the CEA with a high sensitivity of 1.8 μA mL ng-1has a wide linear detection range (LDR) of 0.5-15 ng mL-1 and limit of detection (LOD) as 0.208 ng mL-1. The biosensing characteristics of the fabricated PEDOT/RGO nanocomposite based immunoelectrode reveal that this platform can be efficiently utilized for the fabrication of point of care (POC) device for cancer monitoring.
